Đặc điểm miền chức năng và biến thiên trình tự protein NANB của Pasteurella multocida type D phân lập từ trâu tại tỉnh Đắk Lắk và so sánh với các chủng type B
Tóm tắt
Trong bối cảnh dữ liệu phân tử về Pasteurella multocida type D phân lập từ trâu tại Đắk Lắk còn hạn chế, nghiên cứu này phân tích protein neuraminidase NanB của một chủng type D, ký hiệu EH32, và so sánh với hai chủng type B gồm PM248 và chủng Iran dùng sản xuất vaccine. Trình tự NanB được rút trích từ dữ liệu giải trình tự toàn hệ gen; dự đoán peptide tín hiệu bằng SignalP–5.0, kiến trúc miền được chú giải bằng InterProScan và biến thiên acid amin được đánh giá bằng căn chỉnh đa trình tự. Kết quả cho thấy NanB của cả ba chủng đều mang peptide tín hiệu Sec/SPI với vị trí cắt bảo tồn tại 25/26 (TFG|QI) và có khung kiến trúc miền sialidase ở nửa đầu chuỗi cùng miền β–barrel autotransporter ở C–terminal. Trong phạm vi so sánh với allele NanB của hai chủng type B PM248/Iran, NanB của EH32 có mức độ đồng nhất tổng thể 87,2% tại các vị trí so sánh trực tiếp, trong đó biến thiên tập trung chủ yếu ở vùng passenger/sialidase (PID 76,2%) kèm các indel ngắn, trong khi vùng autotransporter bảo tồn cao (PID 97,7%). Trong phạm vi dữ liệu của nghiên cứu này, các biến thiên ghi nhận ở NanB của chủng EH32 tập trung chủ yếu tại vùng passenger/sialidase, gợi ý đây là vùng cần ưu tiên đánh giá thêm về chức năng sialidase, tính kháng nguyên và thích nghi niêm mạc bằng mô hình hóa cấu trúc 3D hoặc thí nghiệm chức năng.
